pseudonitrosite refs-translation in progress

Antibody has been triing to translate or have traslated several references he has garnered regarding the pseudonitrosite methods, in the interests of expediency antibody will post here the german articles as well as some attempts at translation.

Please feel free to correct or translate any or all of it. Am hoping that with bees collaboaration we will be able to piece togather a good working proceedure.

Also have seen partial refs posted here and there, perhaps they could be compliled here also.

About the utilisation of pseudonitrosites of propenyl-containing phenolic ethers for synthesis of alpha-arylated beta-hydroxyaminopropanols and beta-aminopropanols. New facts for acyl shifts (???)

Vor kurzem konnte der eine von uns zeigen, da die Pseudonitrosite des Isosafrole leicht in die Acetate der entsprechenden  arylierten nitro propanole überzuführen waren.

Recently one of us was able to show that the  pseudonitrosites of isosafrole are easily transformed into the  acetates of the corresponding arylated nitropropanoles

Die Nitro-Propanol ließen sich auf elektrolytischem Wege zu den entsprechenden Hydrolamino Aminoderivaten reduzieren, die nur in Form ihrer Chlorohydrate beständig sind.

The nitropropanoles were easily reduced electrolytically to the corresponding hydroxylamino (?) derivatives, which are stable as the hydrochlorides.

Werden sie aus letzteren in Freiheit gesetzt, so erfolgt eine Acylverschiebung von O zu N, die zu den Derivaten IV und V führt. — Während die Darstellung der entacetylierten Hydrolaminoderivate (VI) — besonders wegen ihrer Empfindlichkeit gegenüber Alkalien — einstweilen nicht gelang, lißen sich die entacetylierten Aminoderivate (VII) leicht fassen.

Wir setzten uns das Ziel, zu diesen Untersuchungen auch das Isoeugenol heranzuziehem. Dies schien uns besonders vom präparativen Standpunkt empfehlenswert, da wir dadurch leicht ein phenol-hydroxyl-haltiges Ephedrinderivat, das p-Oxy-mmethoxy-nor-ephedrin (VIII), aufzubauen gedachten. Da nun das Isoeugenol selbst wegen seiner Sensibilität nicht in das Pseudo-nitrosit überführt werden konnte, bedienten wir uns als Augangssubstanz des Isoeugenol-acetats.

Letzteres konnte durch das a. a. O. angegebene Verfahren in recht befriedigender Ausbeute in das Pseudo-nitrosit (IX)übergeführt werden. Ganz glatt verlief auch die Umwandlung des Pseudo-nitrosites in das Acetate des a(p-Acetoxy-m-methoxy-phenyl)-ß-nitro-propanols (I; Ar=p-Acetoxy-m methoxy-phenyl-). Beide Acetylgruppen der letzeren Verbindug sind recht locker gebuden; schüttelt man nämlich die Substanz mit verdünnter, alkoholisch-wäßriger Kalilauge, so entsteht eine intensiv rote Lösung, aus der sich nach dem Ansäuern — das einen plötzlichen Farbenumschlag ins Hellgelbe verursacht —ß-Nitro-Isoeugenol (X) ausscheidet; letzteres konnte auch durch Einwirkung von alkoholischer Kalilauge auf das Pseudo-nitrosite gewonnen werden). Es wurde schonbei der analogen Bildungsart des ähnlich gebauten ß-Nitroasarone) auf den Mechanismus dieser Reaktion, die zugleich konstitutionsbeweisend für Verbindungen vom Typ 1 ist; hingewiessen.  Beim Isoeugenolderivate gesellt sich diesem Vorgang selbstredend auch noch die hydrolytische Spaltung der p-ständigen Acetoxygruppe zu.

Die Reduktion des Nitro-propanol-derivates I7) wurde auf electrochemischem Weg (Bleikathode) durchgeführt. Von der Strommenge, Stromdichte und Temperature abhängend, konnte die Reduktion derart geregelt werden, daß entweder vorwiegend das Hydroxlamino-oder das Aminoderivat gewonnen wurde. Aus dem mit Natriumacetate abgestumpften bzw. sodaalkalisch gemachten Katholyt wurden beide Reduktionsprodukte als N-Acetylderivate (IV, V 7) isoliert. Die verschiebung des Acetyls von O zu N erfolgt also ebenso, wie bei den analogen Derivaten des Isosafrols und Methyl-isoeugenols. Desglechen konnte dursh Einwirkung von verdünnter Salzäure auch die Rückwanderung des Acetyls bewirkt werden; die O-Acetyl-derivate (II, III)7) ließen sich als Chlorohydrate fassen. Macht man ihre wäßrigen Lösungen sodaalkalisch, so werden weiderum die N-Acetylderivate zurückgebildet. Wir können auf die Erörterung der Konstitutionsbeweise für VerbindungenII-V7) hier verzichten, da sie Analoga der entsprechenden Isosafrole-und Methyl isoeugenol-derivate - deren Konstitution schon früher sichergestellt wurde 1) - verkörpern. Übrigens erhellt die Lage des Acetyls aus Eigenschaften der Verbindungen, die sehr leicht zu beobachten sind (z. B. Löslichkeitsverhältnisse in Säuren und Alkalien, Reduktionsvermögen) und die im Versuchsteil an entsprechenden Stellen angeführt wurden.

Wir konnten feststellen, saß die Acylwanderung von O zu N beim Hydroxylaminoderivat II 7) im strengen Sinne des Wortes nichte augen blicklich vor sich geht und somit die Darstellung verschiedener Nitrone ebenso gelingt, wie dies bei den analogen Derivaten des Isosafrols und Methyl isoeugenols der Fall war 8) Lâßt man nâmlich auf die wäßrige Lösung des Chlorohydrats von II 7) in Genenwart eines fein verteilten Aldehyds Soda einwirken, so setzt sich die in  Freiheit gesetzte Hydroxylaminobase - noch bevor eine Acylverschiebung von O zu N stattfinden könnte - mit dem Aldehyd zum entsprechenden Nitron (XI) um. Es wurde a.a.O. 8) gezeigt, daß die Haftfestigkeit der a-ständigen Acetylgruppe diser Nitrone besonders durch die Substituenten des a-ständigen Aryls, ferner durch die Eigenart des an den Nitroring gekuppelten Restes (R) stark beeienflußt wird. So wird a. B. bei den Nitronen, die sich vom Methyl-isoeugenol ableiten ließen, die Acetylgruppe fallweise (R=Phenyl, 3,4-Dimethoxy-phenyl-,2-Oxy-phenyl) derart aufgelockert, daß sie sich schon durch Einwirkung des schwachen Sodaüberschusses, der bei der Nitronbereitung gebraucht wird, abspaltet; die Reduktion solcher desacetylierten Nitron führte zu Ephedrinabkömmlingen. Beim Derivat des Isoeugenol-acetats (XI) scheint die a-standige Acetylgruppe verhältnismäßig ebenso fest gebunden zu sein, wie bei den analogen Isosafrolderivaten, denn bei der Nitrobildung blieben die acetylierten Produkte stets (R=H-, Phenyl-, 3-nitro-phenyl-) erhalten.

Die Entacetylierung von Verbindung V7) ließ sich bei mäßiger Temperatur mit verdünnter Salzäure leicht durchführen. Auf diese Weise konnte das krystalline Chlorohydrat des a-(p-Oxy-m-methoxy-phenyl)-ß-amino propanols9)(VIII) in befriedigender Ausbeutte gewonnen werden.

Schließlich sei noch kurz der Umwandlung gedacht, die das Pseudo-nitrosite des Isoeugenol-acetats durch siedendes Methanol bzw. salzsäurehaltiges Methanol erleidet. Den Pseudo-nitrositen des Isosafroles und Methyl isoeugenols gleich 10), jedoch viel leichter und schneller, wird auch das Pseudo-nitrosit des Isoeugenol-acetats dursch Kochen mit Methanol vorerst monomerisiert (IX) und darauffolgend tautomer umgelagert; die Entstehung des Nitro-propanol-methyläthers (XIV)7) konnte nicht beobachtet werden. Das bei der tautomeren Umlagerung entstadene Nitroxim (XII), das als krystallines Produkt gefaßt werden konnte, ist in wäßrigen Alkalien sehr leicht löslich; erwartungsgemäß liefert seine alkalische Lösung nach dem Ansäuern das entreprechende Glyoxime-peroxyd (XIII).

Die Tautomerisierungstendenz der Verbindung IX macht sich auch bei der Einwirkung von salzsäurehaltigem Methanol geltend. Es wird nicht - wie beim analogen Asaron- und Methyl-isoeugenol-derivat - der Nitro - propanol - methyläther gebildet XIV), sondern Glyoxime-peroxyd (XIII). Darus kann man nun den Schluß ziehen, daß sich tautomerisierende Wirkung des salzäurehaltigen Methanols nicht nur auf die a-ständige Nitrosogruppe, sondern auch auf die benachbarte Nitrogruppe erstreckt; es entstünde also primär die Säureform des Nitroxims XII, die durch intrmolekulare Wasserabspaltung und eine durch Einwirkung der Salzäure bewirkte Entacetylierung in Verbindung XIV übergehen würde.

Another very helpful bee, (will respect his anonymity though) was kind eneough to trnslate the first part of another reference. Antibody has scanned jpegs of the experimentals that he could also post, but does not know how to go about posting anonymously. And efforts to trasmit via email only causes proxies to crash.

About the usage of Pseudonitosites of Propenyl-containing Phenolethers for the Synthesis of
a-Aryl-b-Hydroxylamino- and b-Aminopropanols
(Part III - Anetholderivates) [1]

In an earlier post in this Journal [2] it has been showed, how y-Nitrosites of Methyl-Isoeugenol, Isoeugenol-Acetat, Isosafrole or Asaron can be used for the formation of
a-Aryl-b-Hydroxylamino- and b-Aminopropanols.
Hydroxylamino-derivates can be used well for the synthesis of Ephedrine-derivates [3], and in spite of the Amino-derivates in rational synthesis of spasmolytic working bases of Isochinolin [4], we decided to expand our field of experiments to the easy-to-get Anethol.
We did this not only for practical reasons, because, as shown earlier, we observed a reversible Acylshift (see below) when experimenting with acetylised Hydroxylamino- and Aminopropanols. To understand that mechanism of the shift of the Acyls our studies should give us more informations.
We transfered our results in synthesis of Nitrosites as in [2] to Anethol and resulted in far better yields as described in other references. The yield of Anethol-y-Nitrosite (I) [5] is significantly lower compared with congenus Phenolether-derivates, because by-products form in this reaction, which are solubly in Ether [6].
Right here we would like to say that we were able to recrystalise the Anethol-y-Nitrosit in Aceton without decomposition in long, clear needles.
Quite smoothly the acetylating conversion of the Nitrosites ran by effect of acetic anhydrid and little phosphoric acid to get a-(4-Methoxy-phenyl)-b-nitro-propanol-acetat (II). This substance is an Oil that we were not able to vacuum distill without decomposition. By the way, constitution-proving is their transformation [7] to b-Nitroanethol (III) by effect of alkalis.
The reduction of the Nitro propanol acetate (II) could be executed by electrolysis in a HCl acidous medium, whereby depending on the quantity of electricity, temperature and condition of the cathode surface we achieved the reduction levels of either the Hydroxylaminos, or the Aminos. From the cathode made blunted with Natriumacetat or soda-alkalinely-made reduction products were seized as N-Acetyl-derivatives (IV, V), since the O-Acetates developed with the reduction (VI, VII) after their release from its Chlorohydrates of a spontaneous acetyl shift of O to N to be subject immediately.
The O-acetates can be synthesided only in form of their salts. Their
Chlorohydratea can be made easily from the N-Acetyl - substances (IV, V) with diluted hydrochloric acid, since then an Ecyl shift of N occurs to O.
The aqueous solution of the chlorhydrate converts them back to their form of  the N-Acetyl-derivates with Soda.
 Since we already reported on the constitution clearing-up of some analogues of connections IV-VII in an earlier post, we don´t need to deal with this question again.
 It is only as much noticed in addition that the position of the acetyl has already been determined - observation was easy, because of easy to watch characteristics, e.g. acid or alkali solubility, reduction ability.
 We will register these briefly in the experiments part.
That the acetyl migration from O to N with the Hydroxylamino-derivate (VI) runs not presently/immediately, we could show here too, which has been known before from equal cases of analogous substances.
If Benzaldehyd is emulsified and if the mixture is made weakly alkaline with soda in an aqueous solution of the chlorohydrates of substance VI, then the reaction is not returning the N-Acetyl-derivate (IV), but the Benzylidennitron of the entacetyliesd Hydroxylamino-base (VIII).
It is thus evident that the condensation reaction takes place in the heterogeneous system with larger rate, as the acetyl shift of O to N in homogeneous reaction systems (i.e. soluted).

The splitting off of the acetyl group, which already took place via effect of little soda surplus, can be regarded as consequence of its untight bonding status. This ability to easy acetylise was observed with numerous Nitron-bondings [8] and the loosening up effect of certain substituents of the aryl, further the radical coupled at the Nitronring, was determined.
The fact that the connection VIII represents an acetylised product could be proven, as follows: if one cooks connection IV with diluted sulfuric acid, then the acetyl group is shifted first by N to O (VI) and then split off; now, if the in such a way received solution of substance X is reacted with Benzaldehyde, then the same Nitron is resulting, which was won also with inclusion of the ecyl migration from substance IV.

The appropriate Propanol amine (IX) could be won by acidous soaping from the n-Acetyl-derivate (V) without difficulty.
This soaping is already caused by a rather moderate intervention, since from a primarily acetyl shift taking place, the O-acetate (VII) results, which can be very easily soaped then.

We could win the chlorhydrate of the amino-base in such a way in very good yield.

Dass die Verbindung VIII ein entacetyliertes Produkt darstellt, konnte folgden bewiesen
werden:

kocht man Verbindung IV mit verdünnter Schwefelsäure, so wird die Acetylgruppe zuerst
von N zu O verschoben (VI) und dann abgespalten;

wird nun die so erhaltene Lösung von Verbindung X mit Benzaldehyd zur Reaktion gebrachz,
so resultiert dasselbe Nitron, das auch mit Einbeziehung der Acylwanderung aus Verbindung IV
gewonnen wurde.

Das entsprechende Propanol-amin (IX) konnte durch saure Verseifung aus dem N-Acetylderivat (V)
ohne Schwierigkeit gewonnen werden.

Diese Verseifung wird schon durch einen ziemlich mässigen Eingriff bewirkt, da durch
eine primär erfolgende Acetylverschiebung das O-Acetat (VII) entsteht, das dann sehr
leicht verseift werden kann.

Das Chlorhydrat der Aminobase konnten wir so in sehr guter Ausbeute gewinnen.
