Hofmann's LSD Synth, Helv.Chim.Acta 26, 944 (1943)

From Helvetica Chemica Acta Vol. 26, 944-65 (1943).
Eine aus 1.0 g D-Isolysergsäure-hydrazid gewonnene ätherische Lösung des Azids versetzte man mit 3.0 cc. Diäthylamin, reinst (Merck), und liess bei 20°C über Nacht stehen. Das Kondensationsprodukt bestand aus einem Gemisch von Lysergsäure- und Isolysergsäure-diäthylamid, das nicht krystallisierte und sich nur durch chromatographische Adsorption zerlegen liess.
Man löste die Substanz in wenig Chloroform, das 1% Alkohol enthielt, adsorbierte mit Aluminiumoxyd nach Brockmann und entwickelte die beiden Zonen mit dem gleichen Lösungsmittel. Der schneller in das Filtrat wandernde Anteil ergab beim Verdampfen einen Rückstand von 0.5 g mit [a]20/D=+84° (c=0.15 in Pyridin), die langsamer wandernde Zone 0.11 g mit [a]20/D=+168° (c=0.15 in Pyridin). Da keine der beiden Fraktionen krystallisierte, wurden sie getrennt nochmals der Adsorptionsanalyse unterworfen, aber diesmal unter Verwendung von Benzol-Aceton 9:1 als Lösungsmittel. Die Fraktion mit [a]20/D=+84° liess sich dabei wieder in zwei Zonen zerlegen, die nun beide krystallisierende Abdampfruckstände lieferten.
Die schneller wandernde Fraktion (0.22 g) krystallisierte aus Chloroform beim Verdünnen mit Äther.  Umkrystallisieren aus Benzol lieferte zugespitzte Prismen mit dem Smp. 80-85°C. Das spezifische Drehvermogen, [a]20/D=+30° (c=0.44 in Pyridin) zeigte, dass D-Lysergsäure-diäthylamid vorliegen musste.
Die langsamer wandernde Zone der Fraktion +84° ergab einen krystallisierten Eindampfrückstand, der identisch war mit der Krystallisation aus der zweiten Adsorptionsanalyse der Fraktion +168°, und der das Amid der Isolysergsäure-Reihe enthielt. Durch Umkrystallisieren aus Aceton, aus dem sich die Substanz in schönen Prismen abscheidet, wurde das D-Isolysergsäure-diäthylamid rein erhalten. Smp.182°C (unter Zersetzung) .

From Helvetica Chemica Acta of volume. 26, 944-65 (1943). D-Isolysergsaeure-hydrazid won ethereal solution of the azide one shifted one from 1.0 g with 3.0 CC. Dia. ethyl amine, reinst (Merck), and left untouched with 20°C uber night. The condensation product consisted of a mixture of Lysergsaeure and Isolysergsaeure diaethylamid, which did not krystallisierte and could only by chromatographische adsorption be divided. One loosened the substance in little chloroform, which contained 1% alcohol, adsorbed with alumina after breaking into man and developed the two zones with the same solvent. The fast moving the portion into the filtrate resulted in in the case of evaporation arrears of 0.5 g with [ a]20/D=+84° (c=0.15 in Pyridin), the more slowly moving zone 0,11 g with [ a]20/D=+168° (c=0.15 in Pyridin). Since none of the two parliamentary groups krystallisierte, they were subjected to the adsorption analysis separately again, but this time using of benzene acetone 9:1 as solvents. The parliamentary group with [ a]20/D=+84° could be divided thereby again into two zones, which supplied now both krystallisierende evaporation jerk conditions. The faster moving parliamentary group (0,22 g) krystallisierte from chloroform when diluting with ether. Umkrystallisieren from benzene supplied course-sharpened prisms with the Smp. 80-85°C. The specific Drehvermogen, [ a]20/D=+30° (c=0.44 in Pyridin) showed that D-Lysergsaeure-diaethylamid had to be present. The more slowly moving zone of the parliamentary group +84° resulted in a krystallisierten eindampfrueckstand, which was identical with the Krystallisation from the second adsorption analysis the parliamentary group +168°, and which contained the amide of the Isolysergsaeure row. By Umkrystallisieren from acetone, from which the substance in beautiful prisms separates, D-Isolysergsaeure-diaethylamid one kept pure. Smp.182°C (under decomposition).

German sentence structure is different than English.Chemical Abstracts vol.38,1499-1501(1944) has the translation. But it omits the final steps of mixing d-isolysergic acid with diethylamine, and its separartion.

Here is another procedure:

Change dimethylamine to 3.0 cc. diethylamine to give D-LSD.
From A.Stoll & A.Hofmann, Helvetica Chemica Acta vol.38,421-33(1955).

1.0g. D-Isolysergsaure-hydrazid wurde in salzsaurer Losung auf ubliche Weise in das Azid ubergefuhrt, dieses nach Freisetzen mit Natriumhydrogencarbonat mit 100 cc. Ather ausgeschuttelt, die Atherlosung 5 Min. mit Pottasche getrocknet und dann im Vakuum bei 10 degrees Badtemperatur zur Trockne eingedampt.  Das hellgelbe, teilweise kristallisierte Azid loste man sogleich bei -10 degrees im Bombenrohr in 10 cc. Dimethylamin.  DAs zugeschmolzene Rohr blieb 1 Std. bei 60 degrees stehen, worauf man die braune Reaktionslosung eindampfte und den Ruckstand and 200 g. Aluminiumoxyd (Brockmann activity III-IV) unter Verwendung von abs. Chloroform als Losungsmittel chromatographierte.  Es bildeten sich zwei im UV-Licht blau leuchtende Zonen.  Die rascher wandernde Zoe enthielt 0.68 g. D-Lysergsaure-dimethylamid.  Die besser haftende Fraktion, die mit Chloroform, das % Alkohol ethielt, eluiert wurde, enthielt das D-isolysergsaure-dimethylamid (0.15 g.).
  
Im Falle des Dimethylamids haftet die Isolysergsaure-Verbingdung demnach fester an der Aluminiumoxydsaule, analog dem Diathylamid und allen bis jetzt dargestellten Isolysergsaure-Derivaten vo sekundaren Aminen, wahrend bei den Amiden von primaren Aminen das Lysergsaure-Derivat ausnahmols besser haftet.

D-LSD-D-tartrate- Biem Zusammengeben von aquivalenten Mengen Saure und Base in 5 Teilen Methanol kristallisiert das salz in langgestreckten kristallosungsmittelhaltigen Prismen von der Zusammensetzung (C2H25ON3)2,C4H6O6,2CH3OH. Smp. 198-20.[a]20/D=+3 degrees (c=1 in wasser).  Beim Troknen im Hochvakuum bei 100 degrees etweicht das Kristallosungsmittel.
 
Die uber das Tartrat gereinigte D-Lysergsaure-diathylamid-Base Zeigte ein spez. Drehvermogen [a]20/D=+17 degrees (c=0.5 in Pyridin) gegenuber dem fruher publizierten Wert(1) von +30 degrees.  Jenes Praparat enthielt noch eie geringe Beimischung von stark rechtsdrehender Isoverbindung.

This is one of the earliest published LSD syntheses. The outline is as follows:

d-Isolysergic acid hydrazide is reacted with sodium nitrite to form d-Isolysergic acid azide, which upon reaction with anhydrous diethylamine forms a mixture of d-isolysergic acid diethylamide and d-lysergic acid diethylamide (LSD). The isomers are separated by column chromatography. The overall yield is ~20%.
