Neurotoxicity

Can somebody try to explain amphetamine neurotoxicity to me?  From what I have read, it seems that free dopamine in the cytoplasm or cytosol induces oxidation of dopamine creating hyperthermia, and permanent damage to certain areas of the brain like the caudate and brain stem.  I frequently read “high dose”, what is a high dose 2.0mg/kg 5.0, 10?  Is high dosage more damaging then chronic use? Are uses with out sleep following some sleep and another 24 hour cycle, less forgiving?  Another part I do not fully understand is glutamate and aspartate attached to amphetamine.  Is this to help facilitate memory or to reduce the risk of neurotoxicity?  Please do not make any guesses, I prefer Rhodium to respond. P.S.  Do any bees notice struggles with spelling and speech with amphetamine? Or struggles with the pronunciation of foreign words?
