Isolation of codeine from solution containing guiacol

The title says it all. Toady hasn't much time left to post, to please excuse the quick post and run.

Anybody know how the two can be separated? Cough mixtures containing little other than H2O, some EtOH, guiacol and codeine are available OTC here. Has anybody ever tried to separate the two? the concentration of codeine is quite low, around 7mg/5ml with 75mg of guiacol.

Dilute with water, basify(NaOH solution), and extract with CH2Cl2/CHCl3. Most of the guiacol will be as the phenolate in the aqeous layer, wash the organic more with dilute NaOH, then water/brine etc. Evaporate to get base, really need vacuum to get rid of solvents..

Depends how much ethanol is there, it may really need to be evaporated first too...Up to you to give it a shot at room temp evaporation perhaps, in a dish to get rid of the bulk of the alcohol present. It will make things easier.

Does the same hold true for dihydrocodeine?

Just happened onto a source of the stuff. Read a paper recently that claimed massive yield improvements on codeine from the 'homebake' pyridine route, using thiols.

I wonder....anyone know if thioketones/thioenones would work the same sort of demethylating magic in such high yields on opioid prodrugs of the codeine type?
